An egg matures inside a small fluid-filled sac in the ovary called a follicle. How follicles grow determines female fertility and the success of in vitro fertilization (IVF).

What they work on

  1. Test whether priming macaques of advanced reproductive age or with poor ovarian response with anti-Müllerian hormone (AMH) improves their response to gonadotropin stimulation and IVF outcomes.
  2. Determine whether adjusting AMH during culture improves follicle growth and egg maturation from vitrified-warmed macaque and human ovarian tissue.
  3. Define how AMH controls cell division and energy production in macaque preantral follicles grown at 5% oxygen, using AMH supplementation or knockdown and analysis of hypoxia-inducible factor, cell cycle and creatine pathways.

More about the research

Anti-Müllerian hormone (AMH), a hormone made by developing follicles, plays a key role. Its effect depends on the follicle's stage. The team's earlier work in rhesus macaques, monkeys whose reproduction closely resembles women's, showed that AMH promotes growth of early (preantral) follicles. At later (antral) stages, it holds back follicle maturation. Timing AMH treatment to the right stage improved follicle growth in the lab and produced eggs (oocytes) able to be fertilized.

Why it matters

Women of advanced reproductive age or with poor response to ovarian stimulation often have limited IVF success. Fertility preservation from frozen ovarian tissue is also still limited. Findings could help optimize IVF protocols and expand fertility preservation options, including for people who freeze ovarian tissue before treatments that harm fertility.
