Nearly every body process, from sleep and metabolism to hormone release, follows a roughly 24-hour circadian rhythm. A brain region called the suprachiasmatic nucleus (SCN) acts as the master clock.

What they work on

  1. Develop flexible Bayesian spatial clustering methods for single-cell spatial transcriptomics data to find how circadian rhythm disruption changes the mix of cell types in mammary tissue.
  2. Identify cell-type-specific spatially varying genes, genes whose activity shifts across the tissue within one cell type, to reveal how circadian disruption affects mammary development at the level of genes and cells.
  3. Build efficient R and Python software packages that implement the new Bayesian spatial methods, to be released and regularly updated for statistics and biomedical researchers.

More about the research

It sends nerve and hormone signals that keep clocks in other tissues, such as the liver and mammary glands, in time. Studies of large populations link circadian rhythm disruption (CRD), such as from shift work or travel across time zones, to metabolic, heart and mood disorders. How CRD changes the mammary gland's tissue environment at the molecular level is not known.

Why it matters

Many people work night shifts or travel across time zones. This work could show whether the resulting circadian disruption affects mammary gland development, and could identify key genes and changes in how cells communicate. The resulting Bayesian methods and free R/Python software could also help other researchers analyze spatial single-cell data in many tissues.
